PuriMag G-streptavidin鏈霉親和素磁珠通過鏈霉親和素和生物素之間的高親和力作用,可以綁定任何生物素標記的分子,如抗體、蛋白質、多肽、DNA等。粒子具有較大的比表面積和很高的捕獲效率。PuriMag G-streptavidin-biotinbiomolecule復合物通過磁分離易于從溶液中分離,因此提供了一個快速、簡潔的磁性納米粒子標記的生物分子的方法。鏈霉親和素磁珠可用于各種下游分離過程:蛋白純化、細胞分離、免疫及分子檢測。鏈霉親和素-生物素(SA-Biotin) 系統具有極高的結合親和力(K =10^-15) , 在生物領域具有廣泛的應用。本產品采用超順磁性微球,粒徑均一、比表面積大、形貌規整,有利于方便、快捷、高效地捕獲目標分子以及實現磁性分離。本產品可配套自動化設備進行高通量操作。

■質量控制
結合能力:每批鏈霉親和素磁珠均用游離生物素測試保證的最小結合能力。
蛋白質的非特異性結合:將2 mg SA磁性顆粒與0.2 mg IgG在200 μl的50 mM磷酸鹽緩沖液(0.15M NaCl,pH7.2)中溫育1小時(15-25℃)。用2×200 ul溫育緩沖液洗滌后,測上清液中的蛋白質濃度(A280)。未檢測到非特定的結合。
無DNA酶活性:將1 μg的DNA底物于37℃下在含有1mg的 SA磁珠的100ml緩沖液M (10mM Tris-HCl,10mM MgCl2,50mM NaCl,1mM DTT,pH7.5)中溫育4小時。未檢測到DNase活性。
無RNA酶活性:將5ug MS2 RNA在37℃下在100ml 10mM Tris-HCl,pH7.5和1mg SA磁性顆粒中溫育4小時。未檢測到RNase活性。
無蛋白酶活性:將10mg的 SA磁珠與casein-resorufin在200μl的0.2M Tris-HCl(20mM CaCl2,pH 7.8)中于37℃溫育30分鐘。 TCA沉淀后,蛋白酶活性與上清液中574nm處的吸光度相關。沒有檢測到蛋白酶活性。
